MacCHESS Advances: Microcrystallography and More

چکیده

Microcrystallography Crystals of proteins and other biological macromolecules never grow very big, due to the “soft” nature of the molecules and the weakness of the forces holding them together. At CHESS and other synchrotron sources, it is routine to deal with crystals that are ~100 μm across, but how often do we need to work with even smaller crystals? Ithaca High School student Sarah Kim, under the direction of MacCHESS staffer Richard Gillilan, surveyed thousands of crystallization attempts (Fig. 1), and concluded that about 50% of initial crystals were in the “micro” category, and that many larger crystals have “good” and “bad” regions and would benefit from use of a microbeam to isolate the better regions.

Protein microcrystallography using synchrotron radiation

The progress in X-ray microbeam applications using synchrotron radiation is beneficial to structure determination from macromolecular microcrystals such as small in meso crystals. However, the high intensity of microbeams causes severe radiation damage, which worsens both the statistical quality of diffraction data and their resolution, and in the worst cases results in the failure of structure...

Structures from Solutions : Biomolecular Small - Angle Solution Scattering at MacCHESS Richard

Protein crystallography continues to be the prime source of high-resolution molecular structural information in biology. Its success depends totally upon the ability of proteins and other biomolecules to form well-ordered crystalline lattices under the right conditions. Many important biological molecules, however, are not well behaved in this regard, a fact that is becoming more apparent as bi...

D3, the new diffractometer for the macromolecular crystallography beamlines of the Swiss Light Source

A new diffractometer for microcrystallography has been developed for the three macromolecular crystallography beamlines of the Swiss Light Source. Building upon and critically extending previous developments realised for the high-resolution endstations of the two undulator beamlines X06SA and X10SA, as well as the super-bend dipole beamline X06DA, the new diffractometer was designed to the foll...
